On which routes will they initially be used? I assume domestic first and then international. Will all 13-14 airframes be based in Australia or will a few be based in Singapore as well?

According to the "Jetmail" emails to which I signed up a while back, their first Revenue flight on the 787 will be on Wednesday 13th of November (Subject to regulatory approval) and is scheduled to be a Melbourne to Gold Coast flight.

Looking at the schedule, they intend to overnight the aircraft at Gold Coast and bring it back to Melbourne the following day.

Subsequent flights include more Melbourne-Gold Coast and some Melbourne - Cairns flights.

18th of December is their intended start date for International flights, with Melbourne to Bali being the first International run for the aircraft.
