If you have done all the obvious checks (Speaker connection, Audio not in mute, rebooted etc) then it's could be a missing or corrupt driver for your sound card....Try to reload the drivers from the CD that came with your computer or go to the sound card manufacturers website and try to find the drivers.

Sometimes, believe it or not, all that's needed is a reboot as sometimes the drivers don't load in properly when Winblows boots up after being turned off.

If your[vista] machine has more than one sound device (might not be apparent), the default sound device can get reset to something that doesn't actually play through the speakers.
Control Panel -> Sound, choose the default device if there's more than one.

Here's one that has me stumped:
Second hand 2.0 Ghz Pentium laptop running XP Home. Re-loaded the sound card drivers. Sound from USB Skype headphones, but not from the 3.5mm jack or the on-board speakers.
I've checked and unchecked every box/button I can find.
