"Squawk" means a transponder code.
The transponder is a bit of equipment on the aircraft which is interrogated by Secondary Surveillance Radar (SSR) on the grond. The transponder then replies with (typically) a 4-digit code.
Later models (Mode "C") also give the aircraft's flight level (based on a pressure of 1013 hPa) and will, in future, provide a lot more information as well (Mode "S").
"Are you squawking?" = "Is your transponder switched on?"
"Squawk 1616 mode C" = "Turn your transponder onto code 1616, with altitude reporting turned on".
