Virgin, only having 747-400 and A340's could also give their FO's first commands on a 747-400 as well.

Like everywhere, 250 hrs, MCC course, self-funded TR and ask everyday for it. They will grant you the LH seat and then you can learn to fly `"0n the job".:}

I don't know of any companies that will give a first command on the 747-400 to a direct entry pilot. However, many airlines allow first commands to pilots who are already working for them as a First Officer. Generally the requirements are about 3000 hours wth a minimum of 500 hours on type, but each airline sets it's own minimum. However as most command oportunities are based on seniority, the people on command courses often have well over this (I got my first command on a 747-200 after 20 years in the RHS and over 10,000 hours on four-engine jets).
I know that some F/O's got their first command on the BA 744's after about 12 years in the airline but I believe that it's longer now (perhaps a BA PPRuNer could confirm).
