If you tick both the DEFO and SO box on the application, they will specify when they call you for interview which position its for. Regarding the sim, the checks are fairly simular, what I've heard is that for DEFO you get very steep turns - 60 degrees bank with stick shaker and they fail an engine downwind after your first approach.
