Separation and the Antonov

There's a prog. on NatGeo right now about the Antonov, that huge cargo plane. Given its size, what kind of effect does that have on the necessary distance between it and anything else flying nearby? How does one calculate something like that, anyway?

Regarding the en-route phase of the flight then there are no special requirements, just standard separation of 3 miles or 5 miles lateraly or 1000ft vertically, depending on which airspace it's in.
On the Approach phase of flight then it will be categorised as a heavy, the same as a 747, 777, etc and have the appropriate distance between itself & the preceding aircraft.

Question is, did he mean the AN-124 or the AN-225?

"Antonov" also covers the AN-2, which is also pretty huge - for a biplane.
