99% of this question is flying a kite, but you never know......
If I wanted to fly somewhere, but didn't want the hassle of going the usual commercail routes etc how would charter a plane for say 25 or even 125 people and going on a flight of say 2,500 miles?
What would the cost be? How is it worked out?

Caution; if you start re-selling the seats or, worse, packaging them with hotels etc, the CAA has a large number of rules, mainly for consumer protection, which you could transgress.
The reality is that usually your group of friends want to stay in the destination for a time, and then come back. So you either have to pay for the aircraft to sit on the ground (the greater part of this this cost is the loss of income, which is a lot) or, more likely, to pay for it to position back to base empty, and then reposition empty to collect the group when the trip is finished. So you have to pay for 2 charters to complete one round trip, unless you can fill the "inside" legs, which is usually impossible.
And don't forget that out of 125 people, 10% will be unable to cough up as promised, 15% will complain that they didn't get what they paid for, 5% will No-Show for excellent reasons and want their money back regardless of the conditions, and 100% will bombard you for 2 months with emails looking for reassurance and information on the best place to leave the dog. The airline will demand full payment 30 days in advance, and then have a 6 hour delay because you're not a big customer and they've got an AOG elsewhere. You will be blamed for that by 100% of the group.

KLM gives a price indication at KLM Royal Dutch Airlines (http://www.klm.com/passage/cityhopper/en/index.html) but there may be additional charges for meals and drinks.

Wow Capot! a travel agent/tour organiser/tour operator that knows about and appreciates demanding Airline Issues and operations :ok: a refreshing change, welcome to the world of Airlines ;).
Well, I did work for one for 11 years, some time ago, and set up another 2 (smallish) ones, one of which is now a big one, but due to my successors' efforts, not mine........the other one is defunct.
